目的探究颅内动脉瘤患者血浆中磷脂酰肌醇蛋白多糖-1(GPC1)的表达特征,比较破裂动脉瘤与未破裂动脉瘤患者血浆中GPC1的表达差异。方法将实验人群分为4组,包括破裂颅内动脉瘤(RIA)和未破裂颅内动脉瘤(UIA)患者各113例,外伤性蛛网膜下腔出血患者50例为疾病对照(DC)组,健康人群43例为健康对照(HC)组。采用酶联免疫吸附法(ELISA)对实验人群进行血浆GPC1含量测定。比较各组血浆GPC1水平,分析其与动脉瘤破裂之间的关系。结果GEO数据库中的基因芯片信息显示,GPC1在动脉瘤壁中的基因表达均显著高于正常颞浅动脉。本研究结果显示,在RIA患者动脉瘤壁中GPC1的蛋白表达水平也明显高于UIA患者。GPC1在RIA患者血浆中蛋白表达明显升高,UIA患者与对照人群中则没有明显变化。Logistics回归分析的结果显示,IA的破裂与GPC1血浆水平(OR:1.774,95%CI=1.146~2.747,P=0.010)密切相关。结论血浆GPC1表达水平是IA破裂的独立危险因素。
ObjectiveTo investigate the expression characteristics of glypican-1(GPC1) in the plasma of patients with intracranial aneurysms, and to compare the differences in the expression of GPC1 in the plasma of patients with ruptured and unruptured aneurysms. MethodsFour experimental groups were set up in this study, including 113 patients with ruptured intracranial aneurysm(RIA) and 113 with unruptured intracranial aneurysm(UIA), 50 patients with traumatic subarachnoid hemorrhage as the disease control(DC) group, and 43 healthy people as the healthy control(HC) group. The plasma GPC1 content of the experimental population was measured by enzyme-linked immunosorbent assay. The plasma GPC1 levels were compared among the groups, and the relationship between them and aneurysm rupture was analyzed, and the ROC curve was used to evaluate the diagnostic value of plasma GPC1 levels on aneurysm rupture. ResultsThe gene chip information in GEO database showed that the gene expression of GPC1 in aneurysm wall was significantly higher than that of superficial temporal artery. This study showed that the protein expression level of GPC1 in aneurysm wall of RIA patients was also significantly higher than that of UIA patients plasma protein expression of GPC1 was significantly increased in RIA patients, but not in UIA patients and control population. The logistic regression analysis showed that rupture of IA was closely associated with GPC1 plasma levels(OR:1.774,95% CI=1.146~2.747,P=0.010). ConclusionPlasma GPC1 expression level is an independent risk factor for IA rupture.
